Restriction digests of the clone give the following sizes (kb): AvaI--4.38; EcoRI/PvuII--2.3, 1.8; NruI--4.2; EcoRI/NruI--3.4, 0.84. The insert contains AvaI, AvaII, StuI(2), HpaI, and SphI sites. Constructed by inserting an XhoI/SstII fragment into vector from which the HindIII/SalI fragment had been removed. The SstIII and HindIII sites were blunt-ended before ligation. |
