An EcoRI/HindIII double digest reveals fragments of 2.5, 1.7, and 1.0 kb from the insert. Restriction digests of the clone give the following sizes (kb): EcoRI/HindIII--3.3, 2.6, 1.5, 0.86; HindIII--4.9, 3.4; EcoRI--7.4, 0.86; PstI--5.6, 1.9, 0.74; BamHI--5.8, 2.4; consistent with deposited DNA. Codominant segregation was demonstrated in 49 3-generation families. MspI also identifies a 2-allele RFLP (2.3, 1.9 kb) but this has not been tested by family studies. Enzyme(s) not detecting polymorphism: BglII, PstI, PvuII, RsaI. |
