Plaque purified on Salk MkHe.  Hemagglutinates a variety of RBC, particularly guinea pig, hamster and human, and also has a CF antigen.  Produces deformity or death in newborn hamsters and neutralizing antibodies in humans, monkeys, hamsters and rats.

      Temperature: 37°C 
Recommendations for Infection:  Plant cells 24-48 hours in advance and infect when cultures are about 70% confluent.   Remove medium and inoculate with a small volume of virus diluted to provide a MOI of about 0.1.  Adsorb 1-2 hours at 37°C in a humidified 5% CO2 atmosphere, rocking every 20-30 minutes to redistribute inoculum.  End adsorption by adding virus growth medium. 
Incubation: Incubate infected culture for 5 days at 37°C temperature in a humidified 5% CO2 atmosphere, until CPE is progressed through 80% of the monolayer.  |
