Animals were immunized with activated mouse macrophages. Spleen cells were fused with Sp2/0-Ag14 myeloma cells. The antibody reacts weakly with resident mouse peritoneal macrophages and strongly with mouse macrophages that have been activated by treatment with Macrophage Activating Factor (MAF) or thioglycollate. It does not react with rat macrophages nor with a panel of mouse macrophage, monocyte or lymphoid cell lines. The antigen is not related to Mac-1, Mac-2 or Mac-3 and does not appear to be strain or sex specific. The antigen is not trypsin sensitive. |

The base medium for this cell line is ATCC Hybri-Care Medium, Catalog No. 46-X. Hybri-Care Medium is supplied as a powder and should be reconstituted in 1 L cell-culture-grade water and supplemented with 1.5 g/L sodium bicarbonate. To make the complete growth medium, add the following components to the base medium: heat-inactivated bovine calf serum to a final concentration of 10%.

Medium Renewal: Every 2 to 3 days Cultures can be maintained by addition or replacement of fresh medium. Start cultures at 2 X 10 exp5 cells/ml and maintain between 1 X 10 exp5 and 1 X 10 exp6 cells/ml. |
