This line does not produce IgM unless stimulated with B Cell Differentiation Factor (BCDF, interleukin-6, interleukin 6, IL-6), however some lots of fetal bovine serum contain levels of endotoxin which may induce a background level of IgM secretion. In order for cells to function optimally in the BCDF assay using IgM secretion, it is essential to pretest fetal bovine serum (FBS) and select lots that are low in endotoxins and do not induce high background levels of IgM secretion. Background secretion should be less than 400 ng/mL.
Tested and found negative for ectromelia virus (mousepox).
Complete Growth Medium
The base medium for this cell line is ATCC-formulated RPMI-1640 Medium, Catalog No. 30-2001. To make the complete growth medium, add the following components to the base medium:
fetal bovine serum to a final concentration of 10%
0.05 mM 2-mercaptoethanol
Subculturing
The cells loosely adhere to the substrate, and subcultures are prepared when the culture is 75% to 90% confluent by striking the flask sharply 3 to 4 times against the palm of the hand. Free floating cells, which accumulate with time in culture, are viable and can be used to initiate new cultures.
Subcultivation Ratio: A subcultivation ratio of 1:3 to 1:5 is recommended
Medium Renewal: Every 2 to 3 days
