Animals were immunized with human cytolytic T lymphocytes. Spleen cells were fused with P3X63Ag8.653 myeloma cells. The beta subunit of LFA-1 is a 95000 dalton polypeptide that is identical to the beta subunits of Mac-1 and the p150,95 antigens. The antibody will inhibit HLA DR mediated T cell cytotoxicity. The depositor states that the cell line should not be maintained in culture continuously for greater than four months or antibody production may be lost. The base medium for this cell line is ATCC Hybri-Care Medium, Catalog No. 46-X. Hybri-Care Medium is supplied as a powder and should be reconstituted in 1 L cell-culture-grade water and supplemented with 1.5 g/L sodium bicarbonate. To make the complete growth medium, add the following components to the base medium: fetal bovine serum to a final concentration of 10%.
