Restriction digests of the clone give the following sizes (kb): PstI--4.4, 0.75; HindIII--5.2; EcoRI--5.2; AvaI--5.2; BamHI--5.2. Detects genomic restriction fragments of the following sizes (kb): owl monkey BamHI--13.0, 3.6. Insert corresponds to bases 332 - 1017 of predicted 1374 mRNA sequence and amino acids 80 - 307. Includes a portion of exon 3 through exon 8. Exon 3-5 and 6-8 specific probes can be generated by HaeII digestion. The 17.2 kb EcoRI genomic fragment is detected by the exon 3 - 5 probe but not the 6 - 8 probe. The 17.2 kb EcoRI fragment is detected in haplotypes A (PGA3, PGA4, PGA5 genes) and B (PGA3, PGA4 genes). The 3.9 kb EcoRI genomic fragment is detected by the exon 6 - 8 probe but not the exon 3 - 5 probe. This fragment is a doublet resulting from two distinct PGA genes. The 13.5 kb EcoRI genomic fragment is detected by the exon 3 - 5 probe but not the exon 6 - 8 probe. This EcoRI fragment is detected in haplotype A which contains the PGA3, PGA4, and PGA5 genes. The 22.0 kb and the 17.8 kb genomic EcoRI fragments are detected by both the exon 3 - 5 and 6 - 8 probes, suggesting the size heterogeneity results from variation at the 5' region of two distinct genes. Recorded as not polymorphic by Collaborative Research, meaning either not polymorphic in 0.8% agarose gels, or difficult to work with. |
