Restriction digests of the clone give the following sizes (kb): EcoRI--2.7, 2.1; BamHI--4.8; PstI--3.3, 1.5; SmaI--4.8; HindIII--4.8. D1S85 has been shown to be tightly linked to D1S86. This probe is a subclone of the 16.41 kb probe VC85. The 2.1 kb sequence in the insert is from the smaller EcoRI allele. The other allele is 2.6 kb. The allele frequencies given were determined in a Caucasian population. Distinct frequencies have been determined in Negro and Bushman (San) populations. This probe detects a 500 bp insertion/deletion polymorphism which can be detected with BglII, StuI, PvuII, PstI, EcoRI, HindIII, and BamHI. Because TaqI and MspI cut within this fragment, allele sizes do not vary by 500 bp. Enzyme(s) not detecting polymorphism: HindII, SstI, AvaII, RsaI, MboI. |
